Defining quaternary RNA architecture of influenza by RNA-RNA seq


Influenza A has segmented genome, comprising 8 negative stranded RNA (vRNA), which offers evolutionary advantages but complicates genomic assembly. Current evidence suggests that influenza vRNPs are organised into a supramolecular complex, but its molecular details remain unknown.

Our goal is to define the quaternary RNA architecture of influenza genomes in cells and virions, by identifying sites of interaction between the 8 vRNPs. To this end, we will use next generation sequencing technology in an original and innovative way to directly and completely define the global vRNA organisation (inter-molecular RNA interactions) and local RNA structure (intra-molecular RNA interactions) of an entire influenza A virus particle.
