Development of template-driven gene editing without DNA breaks

CRISPR Cas technology is largely adopted by scientific communities for applications including modifications in the DNA sequence to nucleic acid detection based diagnosis of infections. However, precise genome editing for correction of hereditary diseases remains a long-standing ambition for this technology. The most recently developed repertoire of CRISPR editors can introduce either single nucleotide changes (Cytosine or Adenine Base editors) or short sequence multi-nucleotide changes (Prime editors), however, their applications are restricted in terms of their editable length and efficiency, respectively. My project aims to develop a novel genome-editing tool that can function without generating double stranded DNA breaks by harnessing the cellular Homologous Recombination Repair machinery.
